Resumen: La dehesa, uno de los sistemas agroforestales más representativos de Europa, presenta un alto valor medioambiental ligado a su equilibrio entre producción y conservación. Por ello, gran parte de los esfuerzos orientados a su preservación se centran en la búsqueda de técnicas que favorezcan su desarrollo sostenible. Con este fin, se consideró el potencial de los hongos endófitos como elemento de biocontrol. Este interés se explica por la relación simbiótica que pueden establecer con una amplia variedad de plantas, asociada normalmente a algún tipo de actividad biológica por parte del hongo, que permitiría estimular el estado general de éstas, así como su resistencia frente a condiciones adversas. Dado que, frecuentemente, dicha bioactividad es consecuencia de la acción de los metabolitos secundarios producidos por cada hongo, estos fueron el objetivo principal del ensayo. Para ello, se procedió al cultivo in vitro de dos hongos, Embellisia leptinellae y otro hongo perteneciente al orden Pleosporales, denominados por los códigos 138 y 234, respectivamente. Dichos hongos fueron seleccionados en base a resultados preliminares prometedores obtenidos de proyectos previos. Una vez llevada a cabo la fermentación, se procedió a la extracción de los metabolitos producidos, con el objetivo de evaluar su bioactividad, centrada en su capacidad antioxidante y antimicrobiana. La primera se estimó en base al método de captura de radicales libres (DPPH), mostrando el hongo E. leptinellae (138) una actividad razonable (IC50: 201,18 μg mL-1). Por su parte, la actividad antimicrobiana se midió a través de la Concentración Mínima Inhibitoria (CMI) de ambos extractos crudos frente al crecimiento de Bacillus subtilis y Botrytis cinerea. En este caso, el hongo 234 (Pleosporales) mostró una inhibición frente a ambos patógenos mayor (37,50 μg mL-1) que el 138 (150 μg mL-1). Ambos hongos presentaron la misma capacidad de inhibir tanto al patógeno de origen bacteriano como al de origen fúngico.
The dehesa, one of the most representative agroforestry systems in Europe, has a great environmental value, linked to its balance between production and preservation. For this reason, much of the efforts aimed to its preservation focus on the search for techniques that favor its sustainable development. For this purpose, the potential of endophytic fungi as a biocontrol tool was considered. This can be explained by the symbiotic relationship that these fungi may establish with a wide range of plants, usually associated with some kind of bioactivity by the fungal partner, which would stimulate the general status of the plant as well as their resistance to adverse conditions. Since this bioactivity is usually the result of the action of the secondary metabolites produced by the fungi, these were the central aim of this work. To do this, 2 fungi, Embellisia leptinellae and another fungus belonging to Pleosporales, designated by the codes 138 and 234 respectively, were cultured in vitro. Both fungi were chosen based on promising preliminary results from previous projects. Once the fermentation was carried out, the metabolites produced were extracted, in order to evaluate their bioactivity through their antioxidant and antimicrobial capacity. The first one was estimated using DPPH radical scavenging assay, showing E. leptinellae (138) a reasonable activity (IC50: 201,18 μg mL-1). On the other hand, the antimicrobial activity was assessed as the Minimum Inhibitory Concentration (MIC) of both crude extracts against the growth of Bacillus subtilis and Botrytis cinerea. In this case, the fungus 234 (Pleosporales) showed an inhibition against both pathogens greater (37,50 μg mL-1) than the fungus 138 (150 μg mL-1). Also, both fungi had the same ability to inhibit both, bacterial and fungal, pathogens.
