dc.description.abstract | The aim of this study was to elucidate if SLC after 24 h storage selects the subpopulation of spermatozoa that better withstands osmotic shock. To test this hypothesis, viability, mitochondrial membrane potential (MMP) and superoxide anion (O2radical dot−) production of uncentrifuged (UC) and single layer centrifugation (SLC) – selected spermatozoa were analyzed following SLC after storage of the semen. An aliquot of the extended ejaculate (100 × 106 spermatozoa/mL) was centrifuged through a single layer of a silane-coated silica based colloid formulation optimized for equine spermatozoa (Androcoll-E large, SLU, Sweden) and the rest was used as control. UC and SLC-sperm samples were subjected to osmotic challenges (75 and 900 mOsm) with a subsequent return to isosmolarity (300 mOsm) using Biggers–Whitten–Whittingham (BWW) medium. Viability and MMP decreased after the different osmotic stress in UC and SLC spermatozoa, and return to isosmolarity did not reverse these effects. O2radical dot− production was enhanced after SLC in all osmolarities tested. Interestingly, the percentage of living spermatozoa showing O2radical dot− production was increased after 900 mOsm stress in UC spermatozoa, this increase being more evident in SLC spermatozoa. Returning spermatozoa to 300 mOsm enhanced this percentage in UC viable cells but not in SLC spermatozoa. The scenario observed for UC spermatozoa shows that O2radical dot− is produced in response to isolated hyperosmolarities and subsequent osmotic excursions. As the viability, MMP and cell volume remained the same between SLC and UC spermatozoa, we conclude that Androcoll-E large is likely selecting a higher percentage of physiologically O2radical dot− producing spermatozoa. | es_ES |
dc.description.abstract | El objetivo de este estudio fue dilucidar si SLC después de 24 h de almacenamiento selecciona la subpoblación de espermatozoides que resiste mejor el shock osmótico. Para probar esta hipótesis, se analizaron la viabilidad, el potencial de membrana mitocondrial (MMP) y la producción de anión superóxido (O2radical punto-) de espermatozoides seleccionados no centrifugados (UC) y de una sola capa (SLC), después del almacenamiento del semen. Se centrifugó una alícuota del eyaculado extendido (100 × 106 espermatozoides/mL) a través de una sola capa de una formulación coloidal a base de sílice recubierta de silano optimizada para espermatozoides equinos (Androcoll-E grande, SLU, Suecia) y el resto se usó como control. . Las muestras de esperma de UC y SLC se sometieron a desafíos osmóticos (75 y 900 mOsm) con un posterior retorno a la isosmolaridad (300 mOsm) utilizando medio Biggers-Whitten-Whittingham (BWW). La viabilidad y la MMP disminuyeron después de los diferentes estrés osmótico en los espermatozoides UC y SLC, y el retorno a la isosmolaridad no revirtió estos efectos. La producción de puntos de radicales O2 mejoró después de SLC en todas las osmolaridades probadas. Curiosamente, el porcentaje de espermatozoides vivos que muestran producción de puntos de radicales O2 aumentó después de un estrés de 900 mOsm en los espermatozoides de UC, siendo este aumento más evidente en los espermatozoides de SLC. Devolver los espermatozoides a 300 mOsm mejoró este porcentaje en células viables de UC, pero no en espermatozoides de SLC. El escenario observado para los espermatozoides de la CU muestra que el punto radical O2 se produce en respuesta a hiperosmolaridades aisladas y excursiones osmóticas posteriores.
