dc.description.abstract | Introducción: la conducta suicida es un problema importante de salud pública en todo el mundo (OMS, 2014). Las personas que realizan tentativas suicidas severas o que utilizan métodos violentos son difíciles de identificar y presentan mayor riesgo de muerte por suicidio. Los marcadores farmacogenéticos pueden ayudar a identificar a este tipo de pacientes. Objetivos: analizar en supervivientes de una tentativa de suicidio el efecto de la capacidad metabólica combinada CYP2D6-CYP2C19 en la severidad y el efecto del polimorfismo ABCB1 en el empleo de un método violento. Métodos: se han analizado las variantes alélicas relacionadas con una actividad enzimática CYP2D6-CYP2C19 nula o incrementada y los SNPs C1236T, G2677T/A y C3435T de ABCB1. Un intento fue severo si la puntuación en la sub-escala “circunstancias objetivables” de SIS (Beck et al., 1974) estaba por encima del percentil 75 y un método fue violento si se empleó ahorcamiento, armas de fuego, etc. (Asberg et al., 1976). Resultados y Discusión: se observó una mayor frecuencia de individuos con tentativa suicida severa entre portadores de una capacidad metabólica CYP2D6-CYP2C19 incrementada. La utilización de un método violento fue más frecuente entre portadores del haplotipo 1236TT-2677TT-3435TT. Estos resultados son consistentes con los publicados previamente (Zackrisson et al., 2010; Peñas-Lledó et al., 2011; Stingl & Viviani, 2011; Peñas-Lledó et al., 2012b; Boiso Moreno et al., 2013). Conclusión: pacientes con una capacidad combinada CYP2D6-CYP2C19 incrementada muestran mayor riesgo de cometer tentativas suicidas severas y el empleo de un método violento es mucho más frecuente entre portadores del haplotipo ABCB1 1236TT-2677TT-3435TT. | es_ES |
dc.description.abstract | Introduction: suicidal behavior is a major public health problem worldwide (OMS, 2014). People who perform severe suicide attempts or who use violent methods are difficult to identify and present a higher risk of death from suicide. The pharmacogenetic markers may be helpful to identify these individuals. Objectives: analyze in survivors of a suicide attempt the effect of combined metabolic capacity CYP2D6-CYP2C19 on the severity of the suicide attempt and the effect of ABCB1 polymorphism on the use of a violent method. Methods: we have analyzed the allelic variants associated with a null or increased CYP2D6-CYP2C19 enzyme activity and SNPs C1236T, G2677T/A, C3435T of the ABCB1 gene. An attempt was severe if the score on the subscale "objetivables circumstances" of SIS (Beck et al., 1974) was above the 75th percentile and a violent method if it was used hanging, firearms, etc. (Asberg et al., 1976). Results and Discussion: a higher frequency of individuals with severe suicide attempt was observed among carriers of a combined high CYP2D6-CYP2C19 metabolic capacity. The use of violent methods was more common among individuals carrying the ABCB1 haplotype 1236TT-2677TT-3435TT. These results are consistent with previously published (Zackrisson et al., 2010; Peñas-Lledo et al., 2011; Stingl & Viviani., 2011; Peñas-Lledo et al., 2012b; Boiso Moreno et al., 2013).
