dc.description.abstract | Aspergillus flavus y Aspergillus parasiticus son especies de moho productoras de aflatoxinas (AF) y pueden crecer en el jamón curado en seco durante el proceso de maduración. En este estudio, se evaluó la influencia de diferentes actividades del agua (aw) y temperaturas en la expresión relativa temporal de tres genes implicados en la biosíntesis de AF y su relación con la producción de AF en un medio a base de jamón curado. En general, los genes reguladores aflR y aflS mostraron patrones de expresión similares, y la expresión del gen estructural aflP fue mucho mayor que la obtenida para los genes aflR y aflS. Respecto a A. flavus, una disminución de aw independientemente de la temperatura provocó un aumento de la expresión de los genes reguladores aflR y aflS. Con respecto a A. parasiticus, los valores de expresión más altos y más bajos de los genes reguladores aflR y aflS se encontraron en 0,95 aw y 0,85 aw, respectivamente. La expresión del gen estructural aflP de ambas especies fue estimulada a baja temperatura y aw. El análisis de PCA indicó que ambas especies toxigénicas mostraron una fuerte correlación entre la expresión relativa de los genes aflR y aflS y la concentración de AF en condiciones que simulan la maduración del jamón curado. Esto sugiere que una detección temprana de la expresión de genes reguladores puede ser un buen indicador de una posible contaminación del jamón curado con AF durante su maduración. | es_ES |
dc.description.abstract | Aspergillus flavus and Aspergillus parasiticus are mould species producers of aflatoxins (AFs) and may grow on dry-cured ham during the ripening process. In this study, the influence of different water activity (aw) and temperatures on the temporal relative expression of three genes involved in AFs biosynthesis and their relationship with AFs production on dry-cured ham-based medium were evaluated. In general, the regulatory aflR and aflS genes showed similar expression patterns, and the expression of the structural aflP gene was much higher than that obtained for aflR and aflS genes. Regarding A. flavus, a decrease of aw regardless of temperature caused an increase of the expression of the regulatory aflR and aflS genes. Concerning A. parasiticus, the highest and lowest expression values of the regulatory aflR and aflS genes were found at 0.95 aw and 0.85 aw, respectively. The expression of the structural aflP gene of both species was stimulated at low temperature and aw. The PCA analysis indicated that both toxigenic species showed a strong correlation between the relative expression of the aflR and aflS genes and the concentration of AFs under conditions which simulate dry-cured ham ripening. This suggests that an early detection of the expression of regulatory genes can be a good indicator of posible AFs contamination of dry-cured ham through ripening. | es_ES |
